春石斛优良品种‘森禾2006’组培快繁体系的建立

摘 要:

本文研究了春石斛‘森禾2006’的拟原球茎(PLBs)诱导、增殖、分化及壮苗和生根, 建立了其组培快繁体系。结果表明: ‘森禾2006’ PLBs最适诱导培养基为1/2MS+TDZ 0.5 mg·L-1+水解酪蛋白2.0 g·L-1; PLBs增殖培养基为1/2MS+水解酪蛋白2.0 g·L-1; PLBs分化培养基为1/2MS+KT 1.0 mg·L-1+NAA 0.1 mg·L-1; 壮苗生根培养基为MS+IBA 0.5 mg·L-1+NAA 0.1 mg·L-1+ 香蕉泥100.0 g·L-1

Establishment of Rapid Propagation System for Elite Variety of Dendrobium nobile Lindl.‘Senhe 2006’by Tissue Culture

Abstract:

To achieve the mass-scale multiplication of Dendrobium nobile ‘Senhe 2006’, the optimal media for the induction, multiplication and differentiation of protocorm-like bodies (PLBs), strengthening and rooting were studied and the rapid propagation system by tissue culture was established. The results showed that 1/2MS+TDZ 0.5 mg·L-1+casein hydrolysate 2.0 g·L-1 was optimal for PLB induction. 1/2MS+casein hydrolysate 2.0 g·L-1 was the most suitable for PLB multiplication. 1/2MS+KT 1.0 mg·L-1+NAA 0.1 mg·L-1 was the most efficient for PLB differentiation. The optimum medium for strengthening and rooting was MS+IBA 0.5 mg·L-1 +NAA 0.1 mg·L-1+banana homogenate 100.0 g·L-1.
